The thromboplastin generation test.

چکیده
Physiological coagulation is dependent on the formation of a powerful thromboplastin within the blood. It is well recognized that coagulation defects, such as that in haemophilia, are due to a failure of the normal thromboplastin mechanism, but lack of knowledge has hitherto prevented any precise or detailed study of these abnormalities. Biggs, Douglas, and Macfarlane (1953a) have shown that three components, normal plasma treated with AI(OH)3, platelets, and normal serum, react to form thromboplastin. Further analysis has shown that two essential substances, antihaemophilic globulin and factor V, occur in the Al(OH)3-treated plasma, while serum contains two factors, factor VII and the Christmas factor, both of which are required for thromboplastin formation (Biggs, Douglas, and Macfarlane, 1953b). These five components-platelets, antihaemophilic globulin, the Christmas factor, and factors V and VII-react together in the presence of CaCl2 to form a labile thromboplastin as powerful as any so far described. A lack of any one of the five produces a coagulation defect associated with abnormal thromboplastin formation. Factors V and VII are usually thought of as

Modified Thrombin Generation Assay: Application To The Analysis Of Immunoglobulin Concentrates
Intravenous immunoglobulins cause thromboembolic events at a frequency of 2-3%. Complications of this nature can be life threatening. Nevertheless, a test for thrombogenicity such as non activated partial thromboplastin time is not mandatory for these drugs. As a result of recent incidents we have performed experiments to evaluate a modified thrombin generation test for its ability to detect mi...
متن کاملAntithromboplastin: the degeneration of intrinsic thromboplastin in normal serum.
It is well known that a powerful thromboplastin is generated during blood coagulation, and a high titre of thromboplastin can be demonstrated in specimens of fresh serum. Its generation may be observed by means of the thromboplastin generation test (Biggs and Douglas, 1953) when a mixture of antihaemophilic globulin, Christmas factor, platelets, factor V, and factor VII is recalcified. If this ...
متن کاملThe degeneration of brain thromboplastin in the presence of normal serum.
It has long been known that the " prothrombin " time of a specimen of plasma to which has been added a brain extract thromboplastin depends on the presence of other factors in addition to prothrombin. Biggs, Douglas, and Macfarlane (1953) have demonstrated that both factor V and factor VII take part in a preliminary reaction with the brain extract to produce an activated thromboplastin. Hardist...
متن کاملDetermination of Biological Activity of Recombinant Reteplase Using Clot Lysis Time and Activated Partial Thromboplastin Time (APTT) Lysis Methods: A Comparative Study
Recombinant plasminogen activator (reteplase) is a third generation thrombolytic agent which has been used on coronary artery thrombosis and acute myocardial infarction. Clot lysis assay is usually considered as a unique method to evaluate biological activity of reteplase. In this study biological activity of reteplase was determined by APTT (activated partial thromboplastin time) lysis method....
متن کاملLaboratory Methodologic Approach in Prolonged Activated Partial Thromboplastin Time Test
The pathological activated partial thromboplastin time test is a data that we can find in all clinical laboratories routinely. The hemato-coagulative clinical case here reported aims to point out how the laboratory can provide a correct diagnosis by methodological rational setting and, consequently, carry out an appropriate therapy, or reassure the patient that the pathological data will not ca...
متن کاملThrombin Generation in Normal Subjects and Cardiac Patients.
• A reliable and sensitive test for both hypoand hypercoagulability would be very desirable for clinical and research purposes. Rapid detection of hypercoagulability would be of value in conditions such as spontaneous clotting fluctuations which occur during stress in patients with congenital or acquired valvular heart diseases < and with previous myocardial infarction. It is difficult to find ...
